A feasible method to extract DNA from the cambium of high-canopy trees: from harvest to assessment Acta Amazonica

ABSTRACT Many tropical trees have high canopies and their leaves are not accessible. Thus, the use of tissue from a more accessible organ (cambium) for DNA extraction may be an alternative for molecular studies. We adapted a feasible methodology for extracting genomic DNA from cambium tissue harvested in the field for the assessment with PCR. We tested three storage conditions (two buffers and a silica gel) and four periods of time after harvest. We used previously described protocols and tested them on three species that occur in Amazonian forests and other biomes: Anadenanthera peregrina var. peregrina, Cedrela fissilis, and Ceiba speciosa. Enhanced Oocyte Activation by Intracytoplasmic Injection of Porcine Spermatozoa Pre-treated with Dithiothreitol OAK

The aim of this study was to confirm the effect of dithiothreitol (DTT)-treated spermatozoa on oocyte activation following intracytoplasmic sperm injection (ICSI). Boar spermatozoa with or without DTT treatment (5 mM, 30 min) were injected into in vitro matured porcine oocytes, and the nuclear phase in presumptive zygotes was observed at 3 h intervals up to 12 h after ICSI. Furthermore, developmental competence of embryos produced by DTT-treated or non-treated spermatozoa was monitored after cultivation in vitro for 144 h. Male and female pronuclear formation rates in the oocytes injected with DTT-treated spermatozoa were significantly (P < 0.05) higher than those in the oocytes injected with non-treated spermatozoa. ABSTRACTStudies that evaluate the physiological and biochemical mechanisms of germination within forest species are needed in order to improve our understanding of such processes. Mercury and dithiothreitol are indicated as important tools in studies that assess the activity of aquaporins during imbibition and germination of seeds. To investigate the alterations caused by mercury inPlathymenia reticulata seedsdifferent doses of mercury were used in the presence and absence of dithiothreitol. Mercury had a dose-dependent effect on the seeds; in the most dilute solutions mercury partially inhibited the imbibition process, whereas in the most concentrated solutions it caused the death of the embryos.
